The Role of Nitric Oxide in Epilepsy.

RATIONALE: Nitric oxide(NO) appears to play a role in epilepsy but available data is controversial.We investigated the role of NO in an acute model.
METHODS: Domoic acid(DA) was injected intraperitoneally(1mg/kg) in 8 week old ICR mice to induce seizures.First group(N:5) was used as control.The second group(N:5)were pretreated with NO donor L- arginine(1000mg/kg) and were given DA 5 mtes later.The third group(N:4) first received NO synthase inhibiter L-nitro arginine methyl ester(L-NAME,10mg/kg)followed 5 mtes later by DA.The animals were observed for 90 mtes after injection for seizures.
RESULTS: All animals in the first group developed hind limb seizures in 20-25 mtes and lasted 45 mtes.In group 2,none of the animals developed seizures.Group 3 mice developed fore and hind limb seizures in 10 min and lasted 60 mtes with one death.
CONCLUSIONS: Nitric oxide precursor L-arginine protects against seizures whereas NO synthase inhibition potentiates DA acid induced seizures.Findings suggest that NO may protect against seizures and its manipulation may have treatment potential.
