Therapeutic Potential of Bifidobacterium infantis on Pilocarpine-Induced Drug-Resistant Epilepsy Rat Model

Rationale: Based on our previous study which found that gut microbiome was significantly altered in patients with drug-resistant epilepsy and higher level of Bifidobacteria was found in patients with fewer seizure frequency, we aimed to explore the preventive effects of Bifidobacterium infantis on drug-resistant epilepsy in a rat model. Methods: Temporal epilepsy model was induced by Pilocarpine. Rats were then administered Bifidobacterium infantis or saline daily. The behavioral manifestations (including seizure frequency, intensity and duration) and electrophysiological features (including sharp wave, spike wave, spike-slow wave and sharp-slow wave) were observed. The expression of p-glycoprotein (P-gp) and multidrug resistance-associated protein 1 (MRP1) in hippocampus was detected by Western-blot and RT-PCR. The gut Bifidobacteria was detected by RT-PCR. Results: Oral administration of Bifidobacterium reduced the incidence of spontaneous epileptic seizures and epileptiform discharge (including sharp wave and spike wave). The expressions of P-gp and MRP1 were suppressed in rats administered Bifidobacterium infantis. Conclusions: Supplementation of Bifidobacteria has therapeutic potential for drug-resistant epilepsy by inhibition of the expression of P-gp and MRP1. Funding: This work has been supported by Sichuan Provincial Health and Family Planning Commission Research Project (No. 18ZD010).
